ناباروری، یک اختلال در سیستم تولید مثلی است که درصد قابل توجهی از علل آن مربوط به مردان می باشد. آسیب های وارده به DNA و عوامل ایجاد کننده ی آن، تاثیر منفی بر نتایج باروری در زوجین دارد. هدف از انجام مطالعه ی حاضر، ارزیابی حیات، DNA فراگمانتاسیون اسپرم و بیان ژن HOTAIR در مردان بارور و نابارور و بررسی ارتباط بین آن ها بود.
روش هانمونه های مایع منی به صورت تصادفی از 25 مرد بارور و 25 مرد نابارور در محدوده ی سنی 55-25 سال که به مرکز باروری و ناباروری حضرت مریم (س) بیمارستان شهید بهشتی اصفهان مراجعه کرده بودند، جمع آوری گردید. ارزیابی حیات اسپرم ها با رنگ آمیزی ایوزین- نگروزین انجام شد. DNA فراگمانتاسیون با Sperm chromatin dispersion test (SCDT) اندازه گیری شد. برای اندازه گیری سطح بیان HOTAIR، از روش Real-time polymerase chain reaction (Real time-PCR) استفاده شد.
یافته هامیانگین بیان HOTAIR در افراد نابارور به طور معنی داری نسبت به گروه بارور کاهش یافت. سطح بیان HOTAIR با آسیب های DNA همبستگی منفی و با حیات اسپرم همبستگی مثبت معنی داری داشت (001/0 > P).
نتیجه گیریکاهش بیان HOTAIR و افزایش آسیب های DNA در افراد نابارور و همبستگی بین آن ها نشان از اهمیت ارزیابی آسیب DNA و تحقیقات پیرامون عوامل ایجاد کننده ی آن ها دارد.

پدیده ی شایع در ارتباط با ورزش های هوازی، آسیب عضلانی و التهاب ناشی از آن می باشد. پژوهش حاضر با هدف بررسی تاثیر دریافت مکمل پروتئین Whey بر التهاب و آسیب عضلانی در ورزشکاران رشته ی بسکتبال انجام شد.
روش هااین مطالعه از نوع کارآزمایی بالینی بود و به مدت 8 هفته بر روی 44 بسکتبالیست 20 تا 30 ساله ی شهر کرمانشاه انجام شد. گروه مداخله، مکمل پروتئین Whey و گروه شاهد، دارونما دریافت نمودند. سطح سرمی آنزیم های آسپارتات آمینوترانسفراز (Aspartate transaminase یا AST)، لاکتات دهیدروژناز (Lactate dehydrogenase یا LDH)، High-sensitivity C-reactive protein (hs-CRP)، اینترلوکین-6 (Interleukin 6 یا IL-6) و کراتین کیناز (Creatine kinase یا CK) و شاخص دردناکی عضلات قبل و پس از مداخله اندازه گیری شد.
یافته هاسطح LDH و CK سرم و شاخص دردناکی عضلات در گروه مداخله نسبت به گروه شاهد کاهش معنی داری را نشان داد، اما سطوح AST و hs-CRP تغییر معنی داری بین دو گروه نداشت. اختلاف معنی داری در سطح IL-6 بین دو گروه مشاهده نشد، اما کاهش سطح IL-6 در گروه مداخله پس از پایان تحقیق وجود داشت. سطوح LDH و CK سرم نیز کاهش معنی داری را در گروه مداخله پس از پایان بررسی نشان داد. همچنین، در گروه مداخله افزایش معنی داری در میزان توده ی عضلانی و کاهش معنی داری در درصد چربی بدن مشاهده گردید.
نتیجه گیریدریافت مکمل پرپروتئین، بر برخی شاخص های آسیب عضلانی ورزشکاران رشته ی بسکتبال تاثیرگذار است.

واکنش پروتئین های قطره ی چربی به تمرین تناوبی، ممکن است منجر به ایجاد شرایطی شود که تشخیص مکانیسم زیربنایی تمرین ورزشی و دیابت نوع دو را فراهم نماید. هدف از انجام پژوهش حاضر، بررسی تاثیر دو شدت متفاوت تمرین تناوبی بر پری لیپین 3 (Perilipin-3 یا PLIN3) بافت چرب احشایی و مقاومت به انسولین رت های نر بود.
روش ها32 سر موش صحرایی نر نژاد ویستار در دو گروه سالم و مبتلا به دیابت قرار گرفتند. سپس موش های مبتلا به دیابت به سه زیرگروه (شاهد مبتلا به دیابت، تمرین تناوبی با شدت بالا و شدت متوسط) و یک گروه شاهد سالم تقسیم شدند تا مطالعه بر روی چهار گروه (هر گروه 8 سر) انجام شود. تمرین در گروه متوسط با 70-65 درصد و در گروه شدت بالا با 90-85 درصد حداکثر اکسیژن مصرفی در طول هشت هفته انجام شد. 48 ساعت پس از آخرین نوبت تمرین، موش ها با تزریق درون صفاقی (Intraperitoneal یا IP) ترکیبی از کتامین و زایلازین بیهوش شدند و بافت چرب دور روده ی آن ها برداشته شد. بیان پروتئین PLIN3 با استفاده از تکنیک Western blot و مقاومت به انسولین نیز به روش Homeostatic Model Assessment-Insulin Resistance (HOMA-IR) صورت گرفت. در نهایت، داده ها با استفاده از آزمون One-way ANOVA و آزمون تعقیبی Tukey تجزیه و تحلیل گردید.
یافته هانتایج آزمون تعقیبی بیانگر کاهش معنی دار PLIN3 و مقاومت به انسولین در گروه تمرین تناوبی با شدت بالا (001/0 = P) و متوسط (001/0 = P) نسبت به گروه شاهد مبتلا به دیابت بود؛ در حالی که تفاوت معنی داری بین گروه های تمرینی با شدت بالا و متوسط وجود نداشت (328/0 = P).
نتیجه گیریتمرینات تناوبی با شدت بالا و متوسط، می تواند منجر به بهبود متابولیسم گلوکز و همچنین، تغییرات چشم گیر در مقادیر بافتی PLIN3 رت های مبتلا به دیابت نوع دو شود.<

Infertility is a disorder of the reproductive system that affects a significant percentage of men. DNA damage and its causative agents negatively affect fertility outcomes. The present study aimed to evaluate the sperm viability, DNA fragmentation, and HOTAIR gene expression in infertile and fertile men, and to investigate the correlation between them.
MethodsSpecimens of semen were obtained randomly from 25 fertile and 25 infertile men, aged between 25-55 years who were referred to Saint Maryam Fertility and Infertility Center of Shahid Beheshti Hospital, Isfahan, Iran. Sperm viability was assessed using Eosin-Nigrosin staining. DNA fragmentation was assessed by sperm chromatin dispersion test (SCDT). The expression of HOTAIR was detected using real-time polymerase chain reaction (real time-PCR).
FindingsHOTAIR gene expression in infertile people was significantly decreased in comparison to fertile group. HOTAIR expression level was negatively correlated with DNA fragmentation and positively correlated with sperm viability (P < 0.001).
ConclusionDecreased HOTAIR expression and increased DNA damage in infertile people and the correlation between them indicating the importance of assessing DNA damage and research on their causative agents.

Muscle damage and inflammation are common phenomena associated with aerobic exercise. The aim of the present study was to evaluate the effect of high-protein supplementation on inflammatory and muscle damage indices in basketball athletes.
MethodsThis clinical trial study was conducted on 44 basketball players aged 20-30 in Kermanshah City, Iran, during an 8-week intervention period. The intervention group was treated with whey protein supplement daily, and the control group received placebo. Serum levels of aspartate transaminase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), high-sensitivity C-reactive protein (hs-CRP), and interleukin 6 (IL-6), as well as muscle pain index were measured </em>before and after intervention.
FindingsLDH, CK, and muscle pain index were significantly decreased in intervention group; however, AST and hs-CRP were not significantly different between the two groups. There were not significant differences in serum IL-6 </em>concentration between two groups, but showed decreases of IL-6 </em>in intervention group at the end of study. Moreover, there was significant decrease in serum LD </em>and CK level in intervention group at the end of study. There was a significant increase in soft lean mass, and a significant decrease in the percent of body fat in the intervention group.
ConclusionThis study showed the efficacy of whey protein intake on some muscle damage markers in basketball athletes.

The reaction of fat droplet proteins to interval exercise may lead to the development of conditions that provide an underlying mechanism for exercise and type 2 diabetes. The aim of this study was to investigate the effect of two different intensities of intermittent exercise on perilipin 3 visceral adipose tissue and insulin resistance in male rats.
Methods32 male Wistar rats were divided into two groups of healthy and diabetic. Then, the diabetic rats were divided into three subgroups of diabetic control and two interval exercise methods (8 in each group). The exercise was performed in the moderate- and high-intensity group with 65-70 and 85-90 percent of the maximum oxygen consumption during eight weeks. 48 hours after the last training session, the mice were anesthetized by intraperitoneal injection of a combination of anesthetic ketamine and xylazine, and adipose tissue around the intestine was removed. The expression level of perilipin 3 protein was assessed by Western blotting, and insulin resistance by the Homeostatic Model Assessment-Insulin Resistance (HOMA-IR) method. The data were analyzed using one-way analysis of variance and Tukey post hoc tests.
ResultsThe post hoc test showed a significant decrease in perilipin 3 and insulin resistance in the high-intensity (P = 0.001) and moderate-intensity (P = 0.001) interval exercise groups compared to the diabetic control group; while there was no significant difference between high- and moderate-intensity interval exercise groups (P = 0.328).
ConclusionHigh-intensity and moderate-intensity intermittent exercise can improve glucose metabolism as well as dramatic changes in the tissue levels of perilipin 3 in type 2 diabetic rats.
